In this publication, Park and colleagues apply genetic circuit design automation (demonstrated previously) with genomically inserted circuits. Landing pads were developed to harbour these circuits and insulation to and from the genome was necessary for the automated circuit design to function properly. For this reason, they developed and characterized double terminators with low readthrough (blocking over 97% of polymerase flux in both directions) to flank these landing pads.
