In this publication, Pinto and colleagues design split sites and characterize a lirary of 34 inteins, 15 of which are sufficiently orthogonal for trans-splicing. The authors have also used these to implement three simultaneous AND gates by splitting ECF sigma-factors. The inteins can also be used to assemble large and repetitive molecules, as shown with spider silk proteins.
